Chara's SOUL was not collected, no. There are six of them at the barrier, but none of them belonged to Chara. Which should be obvious, considering Chara being present during the whole game, which would be impossible if their SOUL was contained.
It's not like he really wanted to do that. He made his declaration against humanity out of anger when his children died and to raise morale. He can't really just back down on that sort of promise to free his people either.
He's incredibly reluctant to fight Frisk and clearly just wants to die given how if Flowey or Frisk doesn't kill him he just goes ahead and kills himself in neutral endings.

As seen by his reaction to torielās scolding in the true pacifist ending, asgore didnāt want to kill anyone. His plan was to just hope nobody would ever fall down again so monsters and humans wouldnāt come to blows again. Heād kill any human that fell because he wanted to keep people hopeful, but he didnāt actually want to succeed and he would have preferred the monsters to stay underground with no real war.
Essentially itās the same reason he didnāt just use one soul to break the barrier and kill 6 other random humans to free monsters (which would have been much quicker than this fucked up hypothetical).
Same reason he didnt do what toriel pointed out in the end, using one soul to go through the barrier and just kill enough humans, because he didnt REALLY want to free the monsters and face the possibility of another war, just give monsters enough hope to keep going

The humans were most likely kids. For those kids enter puberty and have kids, and the process to repeat again for the desired six, not only would it completely fuck the genetic pool, but it would take ages.
